Restylane® Defyne™

The Restylane family of dermal fillers is a range of hyaluronic acid products, including Restylane Silk, Restylane Lyft, Restylane, Restylane Refyne, and Restylane Defyne. They are clear gel formulations of hyaluronic acid, a substance which replicates the hyaluronic acid naturally occurring in the body. Each Restylane product is tailored to a particular purpose.

Restylane Silk is the first FDA-approved filler specifically developed to subtly augment the lips and smooth wrinkles around the mouth. Its gel is significantly smaller, smoother than other Restylane products, creating a softer, more precise outcome. These qualities make Restylane Silk a great option for restoring and perfecting the lips. Moreover, the ultrafine needle enables optimal accuracy and precision when administering the product.

Approved by the FDA, Restylane Lyft is designed to improve volume in the back of the hands, augment cheeks, and address midface contour deficiencies associated with aging. Additionally, it is indicated to treat moderate to severe facial folds, including nasolabial folds (smile lines). Lyft is the only hyaluronic filler approved by the FDA to handle hands and faces, containing 0.3% lidocaine to reduce pain during the procedure.

Restylane is clinically proven to add volume and fullness to the skin, reducing the appearance of moderate to severe facial wrinkles and folds like nasolabial folds. Additionally, Restylane is indicated for lip augmentation.

Restylane Defyne contains a special formulation that smoothly fills in nasolabial folds and marionette lines, while remaining flexible enough to replicate natural facial movements and expressions.

Restylane treatments are permissible only for individuals aged 21 or older.

Dermal fillers are facial injection treatments used to help diminish wrinkles while restoring volume and fullness in the face and hands. There are several types of dermal fillers, one being hyaluronic acid. All Restylane products are transparent hyaluronic acids, which are sugars naturally found in the skin that help provide hydration, fullness and elasticity.

As we age, our skin thins and loses natural fat, leading to wrinkles and loss of facial firmness. This process begins in our 20s, and is affected by both hereditary influences and environmental factors such as sun exposure, smoking, and stress.

WHAT HAPPENS AS WE AGE?

• Hyaluronic acid is a naturally occurring compound in the body, which, coupled with collagen and elastin, gives the skin its suppleness and elasticity.
• Diminishing hyaluronic acid, collagen, and elastin levels with age can cause the skin to become less voluminous and increase wrinkles and sagging.

As people age, the volume of their hands may shrink, making the veins, tendons, and bones more noticeable. This can lead to an aged appearance.

The "cupid's bow" is the iconic double curve in the upper lip, resembling the bow of Cupid, the Roman god of love. The upper lip is thought to be ideal when distinctively formed and symmetrical. Everyone's lip shape and size is unique, and Restylane Silk offers customized shaping and contour precision for silkier, smoother lips and a more natural look.

Patients can enjoy lasting effects with full treatment up to 18 months in nasolabial folds with Restylane, up to 12 months in cheeks with Restylane Lyft, 6 months in lips and perioral rhytids with Restylane Silk, and up to 12 months in facial wrinkles and folds with Restylane Refyne and Defyne.10-13

In clinical trials, the results of Restylane Silk typically lasted up to 6 months post-treatment. As demonstrated by a US clinical trial of mostly female subjects (n=221), 98% noted an observable improvement of both upper and lower lip fullness two weeks after injection, while 77% still experienced an enhancement at the 6-month mark.11

In a trial of 150 participants, evaluators noted that 90% showed an improvement of midface fullness after two months of treatment with Restylane Lyft, and 54% still displayed improvement after 12 months. Satisfaction with midface appearance was high, with 95% of trial members pleased after two months and 73% at 12 months.3 Further, a trial of 89 individuals saw an 86% improvement in the volume of the back of their hands at 3 months; this response rate decreased to 76% at 6 months.3 Evaluators used Merz Hand Grading Scale (MHGS) scores to measure improvement, which was defined as participants having achieved a score of ≥1 at the blinded evaluator's live assessment.

Restylane: In a clinical study treating the lines from the nose to the corners of the mouth (laugh lines), blinded evaluators reported that 95% of subjects improved for up to 18 months when they received an initial Restylane treatment, plus a follow-up treatment approximately 4.5 or 9 months later. Using this Restylane regimen of treatments, blinded evaluators reported similar subject improvement, on average, at 18 months compared to their Week 2 Wrinkle Severity Rating Scale (WSRS) score.10

Restylane Refyne & Restylane Defyne: In a clinical study of subjects treated for laugh lines with Restylane Refyne and Restylane Defyne, investigators reported improvement in the appearance of wrinkles for up to 12 months in the majority of patients. Using the Wrinkle Severity Rating Scale (WSRS), a validated 5-point measure of the size and depth of the wrinkles, with grade 1 defined as absence of wrinkles and grade 5 as extremely deep and long wrinkles, investigators reported that 79% of Restylane Refyne subjects and 77% of Restylane Defyne subjects had at least a 1-grade improvement on the WSRS after 6 weeks. Subjects also performed self-assessments (SSA) of wrinkle severity, with most reporting at least a 1-grade improvement in SSA scores with Restylane Refyne and with Restylane Defyne after 6 weeks.12,14,15

A randomized, evaluator-blinded multicenter trial was conducted to assess the longevity of Restylane's NLF correction. Results indicated Restylane provided 18 months of efficacy with retreatment at four-and-a-half or nine months.

Most Restylane treatments require less than an hour and start to take effect rapidly as it adds volume to even out wrinkles. Composed of hyaluronic acid in a transparent gel, Restylane products mimic the hyaluronic acid found in the body and gradually deteriorate naturally. They all have a unique purpose.

Patients may experience mild side effects such as swelling, redness, pain, bruising, headaches, tenderness, lump formation, itching, and impaired hand function following injection with Restylane Defyne™. These effects generally last under 7 days in nasolabial folds and up to 14-18 days in the lips. A limit of 6.0 mL per treatment is recommended for the treatment of moderate to severe facial wrinkles and folds, and 3.0 mL per hand is advised for volume deficit in the hands. Swelling may be more likely in individuals under 36 years, and bruising more likely in those over 35 years. Be sure to review Important Safety Considerations prior to using any Restylane products.
